It's not a genre I really bother with - and the thing with this kind of question is it's all too easy to slip in "other" films into the category - Amelie for instance, I would not call Rom-Com, as I wouldn't say Big Fish is - but both are v.romantic and both are v.funny.

I was going to suggest Terminator. Waitress meets soldier form the future, they have a misunderstanding but she eventually sees that he is not a threat to her and they go on the run together, hiding from a killer robot from the future. They fall in love and she has his baby. How much more romantic do you want? It was a bit light on the comedy through. :D

As well as some already mentioned (I havent read every post so forgive me if this has already been said) 50 First Dates pulls it off quite well IMO
