Can you simply remove the HD out of a WD MyBook and replace it with a larger on?

As long as it was the same dimensions and had the same connections, I'd say yes.

Only other thought ( as I don't know the product ) would be is there any specific software on there that manages the back ups etc.

It's a Western Digital MtBook essential 250GB, didn't even think if it was sata or ide. It's a few years old, fingers crossed it's sata.
I don't use the software on it for back-ups.
